Abstract   DNA content and proliferating cell nuclear antigen(PCNA) was measured by flow cytometry and immunohistochemical study in 37 patients with transitional cell carcinomas of the bladder who received radical cystectomy. Diploidy was noted in 6(47%) of 13 low grade(grade 1 and 2) tumors, but 7(29%) of Z4 high grade(grade 3) tumors. And the average of positivity of PCNA in low grade was slightly less than high grade, but the difference was not statistically significant. The mean value of positivity of PCNA was 70.1+-13.9% and 76.0+-13.0% for diploid and aneuploid tumors, respectively. This was not statistically significant. The average of pro- liferation index in the group of low positivity of PCNA(positivity of PCNA <= 74%) was 32.6+7.1% and 45.8+-9.0% in the group of high positivity of PCNA(positivity of PCNA>74%). Significant difference was noted between 2 groups, though linear correlation was not noted between the proliferation index and positivity of PCNA. In conclusion, there is no correlation between positivity of PCNA and DNA ploidy, but the proliferation index, reflection of S-phase fraction, has some relation with positivity of PCNA.
